Lion's mane in healthy adults: a useful null pilot
Tested intervention
2 g/day (three vegetable-cellulose capsules) of certified-organic Hericium erinaceus strain M2-102-10 — a combination of mycelial biomass and fruiting body grown on organic whole oats by solid-state fermentation, then dehydrated and milled. This is not evidence about fruiting-body-only powder, a hot-water extract, isolated hericenones/erinacines, or lion's mane generally.
Peer-reviewed · published 26 September and newly indexed 16 October. Design: single-arm pre/post proof-of-concept study; 10 healthy nonsmoking adults aged 18–55 completed 28 days. There was no placebo, no concurrent control and no prospective registry reported.
Outcomes. NIH Toolbox Flanker Inhibitory Control and Attention, Dimensional Change Card Sort, and plasma BDNF. Every pre/post comparison was nonsignificant (all p>0.05). Reported effect sizes (d=0.48, 0.25 and 0.31) are uncontrolled and highly unstable at n=10; they should not be converted into an efficacy signal.
Bias and relevance. Tiny sample, no comparator, practice effects and regression to the mean unresolved. The product was supplied by M2 Ingredients; funding came from an unrestricted International Society of Sports Nutrition grant. Several authors disclosed supplement-industry or consultancy ties. The demographic table also lists an unexplained second group of 10, despite the methods and results describing only one 10-person cohort.
Manuscript verdict
Add a caveat/watch note. Lead with the null outcomes and uncontrolled design; do not change the evidence grade.
